چکیده
Mutation in homologous recombination repair (HRR) pathway is well established ovarian cancers. Multiple trials have shown variable efficacy prostate Mutations other than BRCA1/BRCA2 been recently reported We describe a unique case of an elderly male with metastatic castration-resistant cancer. He responded to hormonal therapy for 6 months but later progressed. Chemotherapy docetaxel produced severe side effects the patient and, thus, was discontinued. Next-generation sequencing performed on tissue sample which selected pathogenic mutation (TIER 1) CHEK2 gene, missense mutation, leading loss function protein. cell cycle regulator and tumor suppressor gene mediating recombinant DNA genetic alterations it makes susceptible newer targeted therapies.

OGG1 DNA Repair Gene Polymorphism As a Biomarker of Oxidative and Genotoxic DNA Damage
Background: Single nucleotide polymorphisms in 8-oxoguanine DNA glycosylase-1 (OGG1) gene modulates DNA repair capacity and functions as one of the first lines of protective mechanisms against 8-hydroxy-2’-deoxyguanosine (8-OHdG) mutagenicity. OGG1-Cys326 gene polymorphism may decrease DNA repair function, causing oxidative stress due to higher oxidative DNA damage. The main purpose of this stu...
متن کاملMtor-Fanconi Anemia DNA Damage Repair Pathway in Cancer.
mTOR is a serine/threonine kinase and plays a critical role in mammalian cell growth, survival, and metabolism. mTOR is present in two cellular complexes: mTORC1 and mTORC2. Dysregulation of the mTOR pathway has been related to tumorigenesis, poor prognosis and/or chemotherapy resistance in a variety of malignancies. Inhibition of mTORC1 by Rapamycin and its analogs has been explored to treat a...
متن کاملRe: Inherited DNA-Repair Gene Mutations in Men with Metastatic Prostate Cancer.
Experts’ summary: In this paper, Pritchard and colleagues tried to ascertain the spectrum and prevalence of germ-line DNA repair gene mutations in metastatic prostate cancer (PCa) by next-generation sequencing. They enrolled a total of 692 patients with metastatic PCa; no selection was based on family history, age, or any knowledge of genetic background. Of the 692 men evaluated, 82 (11.8%) had...
متن کاملInherited DNA-Repair Gene Mutations in Men with Metastatic Prostate Cancer.
BACKGROUND Inherited mutations in DNA-repair genes such as BRCA2 are associated with increased risks of lethal prostate cancer. Although the prevalence of germline mutations in DNA-repair genes among men with localized prostate cancer who are unselected for family predisposition is insufficient to warrant routine testing, the frequency of such mutations in patients with metastatic prostate canc...
متن کاملDefective DNA strand break repair after DNA damage in prostate cancer cells: implications for genetic instability and prostate cancer progression.
Together with cell cycle checkpoint control, DNA repair plays a pivotal role in protecting the genome from endogenous and exogenous DNA damage. Although increased genetic instability has been associated with prostate cancer progression, the relative role of DNA double-strand break repair in malignant versus normal prostate epithelial cells is not known. In this study, we determined the RNA and ...
